Effects of salinity on the growth, physiological and biochemical components of microalga Euchlorocystis marina

Euchlorocystis marina, a new marine species of the genus Euchlorocystis discovered in 2022, has the potential to improve the water quality in mariculture ponds. However, the effects of salinity on the growth, physiology, and biochemical composition of these algae are not well understood. In this study, changes in physiological and biochemical indices such as cell density, photosynthetic pigment, polysaccharide, and lipid content of E. marina under different salinity treatments were analyzed. The results showed that the highest cell density was observed at a salinity of 15‰. The lowest photosynthetic pigment content was observed at a salinity of 60‰, and the highest polysaccharide and lipid content was observed at a salinity of 60‰. These results indicated that lower salinity was more conducive to E. marina reproduction and growth. E. marina can accumulate polysaccharides and lipids in high salinity environments. This study provides new information for understanding the salinity adaptation strategies of E. marina and has practical significance for its development and utilization.

Suillusgrevillei and Suillus luteus promote lead tolerance of Pinus tabulaeformis and biomineralize lead to pyromorphite

Lead (Pb) is a hazardous heavy metal that accumulates in many environments. Phytoremediation of Pb polluted soil is an environmentally friendly method, and a better understanding of mycorrhizal symbiosis under Pb stress can promote its efficiency and application. This study aims to evaluate the impact of two ectomycorrhizal fungi (Suillus grevillei and Suillus luteus) on the performance of Pinus tabulaeformis under Pb stress, and the biomineralization of metallic Pb in vitro. A pot experiment using substrate with 0 and 1,000 mg/kg Pb2+ was conducted to evaluate the growth, photosynthetic pigments, oxidative damage, and Pb accumulation of P. tabulaeformis with or without ectomycorrhizal fungi. In vitro co-cultivation of ectomycorrhizal fungi and Pb shots was used to evaluate Pb biomineralization. The results showed that colonization by the two ectomycorrhizal fungi promoted plant growth, increased the content of photosynthetic pigments, reduced oxidative damage, and caused massive accumulation of Pb in plant roots. The structural characteristics of the Pb secondary minerals formed in the presence of fungi demonstrated significant differences from the minerals formed in the control plates and these minerals were identified as pyromorphite (Pb5(PO4)3Cl). Ectomycorrhizal fungi promoted the performance of P. tabulaeformis under Pb stress and suggested a potential role of mycorrhizal symbiosis in Pb phytoremediation. This observation also represents the first discovery of such Pb biomineralization induced by ectomycorrhizal fungi. Ectomycorrhizal fungi induced Pb biomineralization is also relevant to the phytostabilization and new approaches in the bioremediation of polluted environments.

Enhancing the phytoextraction efficiency of heavy metals in acidic and alkaline soils by Sedum alfredii Hance: A study on the synergistic effect of plant growth regulator and plant growth-promoting bacteria

Plant growth regulators (PGR) and plant growth-promoting bacteria (PGPB) have the potential in phytoremediation of heavy metals (HMs) contaminated soils. However, their sole application may not yield the optimal results, thus necessitating the combined application. The present study aimed to enhance the phytoremediation efficiency of Sedum alfredii Hance (S. alfredii) in acidic and alkaline soils through the combination of PGR (Brassinolide, BR) and PGPB (Pseudomonas fluorescens, P. fluorescens). The combination of BR and P. fluorescens (BRB treatment) effectively increased the removal efficiency of S. alfredii for Cd, Pb, and Zn by 355.2 and 155.3 %, 470.1 and 128.9 %, and 408.4 and 209.6 %, in acidic and alkaline soils, respectively. Moreover, BRB treatment led to a substantial increase in photosynthetic pigments contents and antioxidant enzymes activities, resulting in a remarkable increase in biomass (86.71 and 47.22 %) and dry mass (101.49 and 42.29 %) of plants grown in acidic and alkaline soils, respectively. Similarly, BRB treatment significantly elevated the Cd (109.4 and 71.36 %), Pb (174.9 and 48.03 %), and Zn levels (142.8 and 104.3 %) in S. alfredii shoots, along with cumulative accumulation of Cd (122.7 and 79.47 %), Pb (183.8 and 60.49 %), and Zn (150.7 and 117.9 %), respectively. In addition, the BRB treatment lowered the soil pH and DTPA-HMs contents, while augmenting soil enzymatic activities, thereby contributing soil microecology and facilitating the HMs absorption and translocation by S. alfredii to over-ground tissues. Furthermore, the evaluation of microbial community structure in phyllosphere and rhizosphere after remediation revealed the shift in microbial abundance. The combined treatment altered the principal effects on S. alfredii HMs accumulation from bacterial diversity to the soil HMs availability. In summary, our findings demonstrated that synergistic application of BR and P. fluorescens represents a viable approach to strengthen the phytoextraction efficacy of S. alfredii in varying soils.

Plant-microbe involvement: How manganese achieves harmonious nitrogen-removal and carbon-reduction in constructed wetlands

Carbon deficits in inflow frequently lead to inefficient nitrogen removal in constructed wetlands (CWs) treating tailwater. Solid carbon sources, commonly employed to enhance denitrification in CWs, increase carbon emissions. In this study, MnO2 was incorporated into polycaprolactone substrates within CWs, significantly enhancing NH4+-N and NO3–-N removal efficiencies by 48.26–59.78 % and 96.84–137.23 %, respectively. These improvements were attributed to enriched nitrogen-removal-related enzymes and increased plant absorption. Under high nitrogen loads (9.55 ± 0.34 g/m3/d), emissions of greenhouse gases (CO2, CH4, and N2O) decreased by 147.23–202.51 %, 14.53–86.76 %, and 63.36–87.36 %, respectively. N2O emissions were reduced through bolstered microbial nitrogen removal pathways by polycaprolactone and MnO2. CH4 accumulation was mitigated by the increased methanotrophs and dampened methanogenesis, modulated by manganese. Additionally, manganese-induced increases in photosynthetic pigment contents (21.28–64.65 %) fostered CO2 sequestration through plant photosynthesis. This research provides innovative perspectives on enhancing nitrogen removal and reducing greenhouse gas emissions in constructed wetlands with polymeric substrates.

Cultivar-specific regulation of antioxidant enzyme activity and stomatal closure confer tolerance of wheat to elevated ozone: A two-year open-field study with five cultivars

Elevated concentrations of tropospheric ozone (O3) pose a significant threat to food production in many regions of the world. We conducted a two-year experiment with five winter wheat (Triticum aestivum L.) cultivars, Yangmai16 (Y16), Yangmai23 (Y23), Zhenmai12 (Z12), Yangmai29 (Y29), and Yangmai30 (Y30), to assess photosynthetic traits, lipid peroxidation, and antioxidant systems under ambient (A-O3) or elevated (E-O3; 1.5 times A-O3) O3 in a Free-Air O3-Concentration Enrichment system (FACE). E-O3 accelerated leaf senescence, manifested by marked reductions in photosynthetic rate, stomatal conductance, total antioxidant capacity, and photosynthetic pigment content, accompanied by altered antioxidant enzyme activity and a significant increase in malondialdehyde content (an indicator of lipid peroxidation). Most parameters showed significant inter-cultivar differences, as well as interactions between O3 and cultivar. The response of the antioxidant system in wheat flag leaves was influenced by O3 exposure levels. The response of antioxidant enzymes at the grain filling stage and the timely closure of stomata conferred tolerance to O3. This tolerance was associated with higher antioxidant enzyme activity at the anthesis stage but was not related to the size of stomata. This study provides a crucial theoretical foundation for further breeding O3-tolerant cultivars.

Improvement of morphophysiological and anatomical attributes of plants under abiotic stress conditions using plant growth-promoting bacteria and safety treatments.

Drought and salinity are the major abiotic stress factors negatively affecting the morphophysiological, biochemical, and anatomical characteristics of numerous plant species worldwide. The detrimental effects of these environmental factors can be seen in leaf and stem anatomical structures including the decrease in thickness of cell walls, palisade and spongy tissue, phloem and xylem tissue. Also, the disintegration of grana staking, and an increase in the size of mitochondria were observed under salinity and drought conditions. Drought and salt stresses can significantly decrease plant height, number of leaves and branches, leaf area, fresh and dry weight, or plant relative water content (RWC%) and concentration of photosynthetic pigments. On the other hand, stress-induced lipid peroxidation and malondialdehyde (MDA) production, electrolyte leakage (EL%), and production of reactive oxygen species (ROS) can increase under salinity and drought conditions. Antioxidant defense systems such as catalase, peroxidase, glutathione reductase, ascorbic acid, and gamma-aminobutyric acid are essential components under drought and salt stresses to protect the plant organelles from oxidative damage caused by ROS. The application of safe and eco-friendly treatments is a very important strategy to overcome the adverse effects of drought and salinity on the growth characteristics and yield of plants. It is shown that treatments with plant growth-promoting bacteria (PGPB) can improve morphoanatomical characteristics under salinity and drought stress. It is also shown that yeast extract, mannitol, proline, melatonin, silicon, chitosan, α-Tocopherols (vitamin E), and biochar alleviate the negative effects of drought and salinity stresses through the ROS scavenging resulting in the improvement of plant attributes and yield of the stressed plants. This review discusses the role of safety and eco-friendly treatments in alleviating the harmful effects of salinity and drought associated with the improvement of the anatomical, morphophysiological, and biochemical features in plants.

Alpha lipoic acid mitigates adverse impacts of drought stress on growth and yield of mungbean: photosynthetic pigments, and antioxidative defense mechanism.

Exogenous use of potential organic compounds through different modes is a promising strategy for the induction of water stress tolerance in crop plants for better yield. The present study aimed to explore the potential role of alpha-lipoic acid (ALA) in inducing water stress tolerance in mungbean lines when applied exogenously through various modes. The experiment was conducted in a field with a split-plot arrangement, having three replicates for each treatment. Two irrigation regimes, including normal and reduced irrigation, were applied. The plants allocated to reduced irrigation were watered only at the reproductive stage. Three levels of ALA (0, 0.1, 0.15 mM) were applied through different modes (seed priming, foliar or priming+foliar). ALA treatment through different modes manifested higher growth under reduced irrigation (water stress) and normal irrigation. Compared to the other two modes, the application of ALA as seed priming was found more effective in ameliorating the adverse impacts of water stress on growth and yield associated with their better content of leaf photosynthetic pigments, maintenance of plant water relations, levels of non-enzymatic antioxidants, improved activities of enzymatic antioxidants, and decreased lipid peroxidation and H2O2 levels. The maximum increase in shoot fresh weight (29% and 28%), shoot dry weight (27% and 24%), 100-grain weight (24% and 23%) and total grain yield (20% and 21%) in water-stressed mungbean plants of line 16003 and 16004, respectively, was recorded due to ALA seed priming than other modes of applications. Conclusively, 0.1 and 0.15 mM levels of ALA as seed priming were found to reduce the adverse impact of water stress on mungbean yield that was associated with improved physio-biochemical mechanisms. The findings of the study will be helpful for the agriculturalists working in arid and semi-arid regions to obtain a better yield of mungbean that will be helpful to fulfill the food demand in those areas to some extent.

Assessment of the effect of organic fertilizers on growth and biochemical parameters of <i>Lepidium sativum</i> and <i>Raphanus sativus</i>

Relevance. The article presents the results of studying the effect of organic fertilizers of various origins on vegetable crops of radish and watercress. The relevance of the study is related to the need to find the most effective preparations based on humic acids from a wide list offered for use in crop production.Methods. The research was carried out in a model experiment in 2022–2023 with humic fertilizers of geological (“Humate K”) and biological (“AgroVerm” and “Humate K”) origin. The study of the effect of meliorants was carried out on vegetable crops: radish of the Heat variety and watercress of the Moscow Greenhouse variety. The data obtained allow us to conclude that the application of organic fertilizers significantly increases germination, stimulates the growth and development of crops. The effect of humic preparations significantly increased growth indicators and the content of photosynthetic pigments in the green mass of plants with control. For the watercress culture, the highest yield values (1.46 kg /m2) were noted against the background of the introduction of the drug “Humate K”, and for radish, the maximum yield value (2.8 kg/ m2) was observed in the variant of using the effluent Samorod.Results. The results of the study confirmed the high efficiency of using humic fertilizers in conditions of low soil fertility and revealed the selective sensitivity of vegetable crops to humic preparations of various origins.

Plastic Litter Emits the Foraging Infochemical Dimethyl Sulfide after Submersion in Freshwater Rivers.

Plastic pollution is widespread throughout aquatic environments globally, with many organisms known to interact with and ingest plastic. In marine environments, microbial biofilms that form on plastic surfaces can produce the odorous compound dimethyl sulfide (DMS), which is a known foraging cue. This has been shown to increase the ingestion of plastic by some invertebrates and therefore act as a biological factor which influences the risks of plastic to marine ecosystems. In freshwater however, the production of DMS has been largely overlooked, despite the known sensitivity of some freshwater species to this compound. To address this gap, the present study analyzed the production of DMS by biofilms which formed on low-density polyethylene and polylactic acid films after 3 and 6 weeks of submersion in either a rural or an urban United Kingdom river. Using gas chromatography-mass spectrometry, the production of DMS by these biofilms was consistently identified. The amount of DMS produced varied significantly across river locations and materials, with surfaces in the urban river generally producing a stronger signal and plastics producing up to seven times more DMS than glass control surfaces. Analysis of biofilm weight and photosynthetic pigment content indicated differences in biofilm composition across conditions and suggested that DMS production was largely driven by nonphotosynthetic taxa. For the first time this work has documented the production of DMS by plastic litter after submersion in freshwater rivers. Further work is now needed to determine if, as seen in marine systems, this production of DMS can encourage the interaction of freshwater organisms with plastic litter and therefore operate as a biological risk factor in the impacts of plastic on freshwater environments. The influence of exogenous phytohormones and zinc sulfate on the morphophysiological characteristics of Salvinia natans (Salviniaceae)

Hydrophytic ferns of the family Salviniaceae, such as those of the genus Salvinia, hold promise for application in water purification and bioassessment. The Salvinia species are known for their rapid growth, adaptability, resistance to adverse environmental conditions, and also their ability to adsorb pollutants. The manipulation of plant growth and development through the application of exogenous phytohormones is an effective technique. In laboratory settings, we investigated the impact of exogenous phytohormones and zinc sulfate on the morphophysiological characteristics of young sporophytes of Salvinia natans. During the phase of intensive growth, the addition of exogenous abscisic acid (ABA) at a concentration of 10−5 M to the growth medium inhibited the accumulation of biomass in S. natans sporophytes. However, gibberellic acid (GA3) and cytokinins (kinetin or zeatin) at a concentration of 10−6 M, along with indole-3-acetic acid (IAA) at a concentration of 10−5 M, induced a slight increase in biomass. In sporophytes of S. natans grown in a medium containing varying concentration of zinc sulfate (228, 114, 57, 28.5, and 14.25 mg of zinc per liter of water), sings of chlorosis in floating fronds were observed as early as the second day of cultivation, followed by symptoms of necrosis. There was a decrease in the content of photosynthetic pigments. Sporophyte biomass decreased, as zinc concentration increased, ranging from 9.6% to 51%. Simultaneous application of phytohormones with the highest concentration of zinc sulfate partially mitigated the adverse effects of the metal, reducing biomass degradation and sustaining sporophyte viability. Qualitative alterations in the fern phenotype were observed, highlighting the phytotoxicity of excessive zinc concentrations. This underscores the potential use of S. natans as a bioindicator for water pollution.

Model of decomposition of the native absorption spectrum of Porphyridium purpureum culture

A model of the native absorption spectrum of the red seaweed Porphyridium purpureum culture was developed in this work. The mathematical model of each pigment is the sum of Gaussian curves. To level the light scattering, the spectra of the culture were recorded on a spectrophotometer with an integrating sphere. To verify the model, a series of parallel measurements of photosynthetic pigment concentrations using standard biochemical methods and the Gaussian curve method were performed. It was shown that the proposed model with sufficient accuracy makes it possible to determine the concentration of the main photosynthetic pigments of Porphyridium purpureum culture without interfering with its growth processes.

Enhancing sweet potato (Ipomoea batatas) resilience grown in cadmium-contaminated saline soil: a synergistic approach using Moringa leaf extract and effective microorganisms application.

Raising soil contamination with cadmium (Cd2+) and salinization necessitates the development of green approaches using bio-elicitors to ensure sustainable crop production and mitigate the detrimental health impacts. Two field trials were carried out to study the individual and combined effects of foliage spraying of Moringa leaf extract (MLE) and soil application of effective microorganisms (EMs) on the physio-biochemical, osmolytes, antioxidants, and performance of sweet potato grown in Cd2+-contaminated salty soil (Cd2+  = 17.42mgkg-1 soil and soil salinity ECe = 7.42 dS m-1). Application of MLE, EMs, or MLE plus EMs significantly reduced the accumulation of Cd2+ in roots by 55.6%, 50.0%, or 68.1% and in leaves by 31.4%, 27.6%, or 38.0%, respectively, compared to the control. Co-application of MLE and EMs reduced Na+ concentration while substantially raising N, P, K+, and Ca2+ acquisition in the leaves. MLE and EMs-treated plants exhibited higher concentrations of total soluble sugar by 69.6%, free proline by 47.7%, total free amino acids by 29.0%, and protein by 125.7% compared to the control. The enzymatic (SOD, APX, GR, and CAT) and non-enzymatic (phenolic acids, GSH, and AsA) antioxidants increased in plants treated with MLE and/or EMs application. Applying MLE and/or EMs increased the leaf photosynthetic pigment contents, membrane stability, relative water content, water productivity, growth traits, and tuber yield of Cd2+ and salt-stressed sweet potato. Consequently, the integrative application of MLE and EMs achieved the best results exceeding the single treatments recommended in future application to sweet potato in saline soil contaminated with Cd2+.

Non-proteinogenic amino acids mitigate oxidative stress and enhance the resistance of common bean plants against Sclerotinia sclerotiorum.

White mold, caused by the necrotrophic fungus Sclerotinia sclerotiorum, is a challenging disease to common bean cultivation worldwide. In the current study, two non-proteinogenic amino acids (NPAAs), γ-aminobutyric acid (GABA) and ß-alanine, were suggested as innovative environmentally acceptable alternatives for more sustainable management of white mold disease. In vitro, GABA and ß-alanine individually demonstrated potent dose-dependent fungistatic activity and effectively impeded the radial growth and development of S. sclerotiorum mycelium. Moreover, the application of GABA or ß-alanine as a seed treatment followed by three root drench applications efficiently decreased the disease severity, stimulated plant growth, and boosted the content of photosynthetic pigments of treated S. sclerotiorum-infected plants. Furthermore, although higher levels of hydrogen peroxide (H2O2), superoxide anion (O2 •-), and malondialdehyde (MDA) indicated that S. sclerotiorum infection had markedly triggered oxidative stress in infected bean plants, the exogenous application of both NPAAs significantly reduced the levels of the three studied oxidative stress indicators. Additionally, the application of GABA and ß-alanine increased the levels of both non-enzymatic (total soluble phenolics and flavonoids), as well as enzymatic (catalase [CAT], peroxidases [POX], and polyphenol oxidase [PPO]) antioxidants in the leaves of S. sclerotiorum-infected plants and improved their scavenging activity and antioxidant efficiency. Applications of GABA and ß-alanine also raised the proline and total amino acid content of infected bean plants. Lastly, the application of both NPAAs upregulated the three antioxidant-related genes PvCAT1, PvCuZnSOD1, and PvGR. Collectively, the fungistatic activity of NPAAs, coupled with their ability to alleviate oxidative stress, enhance antioxidant defenses, and stimulate plant growth, establishes them as promising eco-friendly alternatives for white mold disease management for sustainable bean production.

The effects of cadmium on selected oxidative stress parameters and the content of photosynthetic pigments in cucumber Cucumis sativus L.

BackgroundEnvironmental pollution by cadmium (Cd) is currently a common problem in many countries, especially in highly industrialised areas. Cd present in the soil can be absorbed by plants through the root system. AimThe aim of the present study was to investigate the effects of cadmium on the metabolic activity of cucumber plants (Cucumis sativus L.) and the accumulation and distribution of Cd in the organs of the plants. MethodsCucumber seeds (3 g) were exposed to 0.76, 1.58 or 4.17 mg Cd/L (applied as CdCl2 solutions). The activity of selected antioxidant enzymes - glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and catalase (CAT), lipid peroxidation and the content of photosynthetic pigments were determined in 6-week-old cucumber plants. In addition, intake of Cd has been determined by flame atomic absorption spectrometry (F-AAS). ResultsThe results show that the applied cadmium concentrations affected the activity of antioxidant enzymes. An increase in CAT activity and a decrease in SOD activity were observed in all cucumber organs analysed. GSH-Px activity increased in the roots and stems. Surprisingly, GSH-Px activity decreased in the leaves. The level of lipid peroxidation was usually unchanged (the only one statistically significant change was a decrease in the concentration of malondialdehyde in the leaves which was observed after exposure to the highest Cd concentration). The applied Cd concentrations had no effect on the content of photosynthetic pigments. The highest cadmium content was found in the roots of cucumber plants. Cd tends to accumulate in the roots and a small amount was translocated to the stems and leaves, which was confirmed with the translocation factor (TF). ConclusionsThe results indicate that the range of cadmium concentrations used, corresponding to the level of environmental pollution recorded in Europe, effectively activates the antioxidant enzyme system, without intensifying lipid peroxidation or reducing the content of photosynthetic pigments.

Multi-biomarker response of cyanobacteria Synechocystis salina and Microcystis aeruginosa to diclofenac

The cyanobacterial response to pharmaceuticals is less frequently investigated compared to green algae. Pharmaceuticals can influence not only the growth rate of cyanobacteria culture, but can also cause changes at the cellular level. The effect of diclofenac (DCF) as one of the for cyanobacteria has been rarely tested, and DCF has never been applied with cellular biomarkers. The aim of this work was to test the response of two unicellular cyanobacteria (Synechocystis salina and Microcystis aeruginosa) toward DCF (100 mg L−1) under photoautotrophic growth conditions. Such endpoints were analyzed as cells number, DCF uptake, the change in concentrations of photosynthetic pigments, the production of toxins, and chlorophyll a in vivo fluorescence. It was noted that during a 96 h exposure, cell proliferation was not impacted. Nevertheless, a biochemical response was observed. The increased production of microcystin was noted for M. aeruginosa. Due to the negligible absorption of DCF into cells, it is possible that the biochemical changes are induced by an external signal. The application of non-standard biomarkers demonstrates the effect of DCF on microorganism metabolism without a corresponding effect on biomass. The high resistance of cyanobacteria to DCF and the stimulating effect of DCF on the secretion of toxins raise concerns for environment biodiversity.

Effects of four antibiotics on the photosynthetic light reactions in the green alga Chlorella pyrenoidosa

Antibiotics are ubiquitously present in aquatic environments, posing a serious ecological risk to aquatic ecosystems. However, the effects of antibiotics on the photosynthetic light reactions of freshwater algae and the underlying mechanisms are relatively less understood. In this study, the effects of 4 representative antibiotics (clarithromycin, enrofloxacin, tetracycline, and sulfamethazine) on a freshwater alga (Chlorella pyrenoidosa) and the associated mechanisms, primarily focusing on key regulators of the photosynthetic light reactions, were evaluated. Algae were exposed to different concentrations of clarithromycin (0.0–0.3 mg/L), enrofloxacin (0.0–30.0 mg/L), tetracycline (0.0–10.0 mg/L), and sulfamethazine (0.0–50.0 mg/L) for 7 days. The results showed that the 4 antibiotics inhibited the growth, the photosynthetic pigment contents, and the activity of antioxidant enzymes. In addition, exposure to clarithromycin caused a 118.4 % increase in malondialdehyde (MDA) levels at 0.3 mg/L. Furthermore, the transcripts of genes for the adenosine triphosphate (ATP) - dependent chloroplast proteases (ftsH and clpP), genes in photosystem II (psbA, psbB, and psbC), genes related to ATP synthase (atpA, atpB, and atpH), and petA (related to cytochrome b6/f complex) were altered by clarithromycin. This study contributes to a better understanding of the risk of antibiotics on primary producers in aquatic environment.

Regulation of photosynthetic function and reactive oxygen species metabolism in sugar beet (Beta vulgaris L.) cultivars under waterlogging stress and associated tolerance mechanisms

Sugar beet (Beta vulgaris L.) is an economically important sugar crop worldwide that is susceptible to sudden waterlogging stress during seedling cultivation, which poses a major threat to sugar beet development and production. Our understanding of the physiological basis of waterlogging tolerance in sugar beet is limited. To investigate the photosynthetic adaptation strategies of sugar beet to waterlogging stress conditions, the tolerant cultivar KUHN1260 (KU) and sensitive cultivar SV1433 (SV) were grown under waterlogging stress, and their photosynthetic function and reactive oxygen species (ROS) metabolism were assessed. Our results showed that waterlogging stress significantly reduced the photosynthetic pigment content, rubisco activity, and expression level of the photosynthetic enzyme genes SvRuBP, SvGAPDH, and SvPRK, gas exchange parameters, and chlorophyll fluorescence parameters, induced damage to the ultrastructure of the chloroplast of the two sugar beet cultivars, inhibited the photosynthetic carbon assimilation capacity of sugar beet leaves, damaged the structural stability of photosystem II (PSII), and disturbed the equilibrium between electrons at the acceptor and donor sides of PSII, which was the result of stomatal and non-stomatal limiting factors. Moreover, the level of ROS, H2O2, and O2▪−, antioxidant enzyme activity, and gene expression levels in the leaves of the two sugar beet cultivars increased over time under waterlogging stress; ROS accumulation was lower and antioxidant enzyme activities and gene expression levels were higher in the waterlogging-tolerant cultivar (KU) than the waterlogging-sensitive cultivar (SV). In sum, these responses in the more tolerant cultivars are associated with their resistance to waterlogging stress. Our findings will aid the breeding of waterlogging-tolerant sugar beet cultivars.

Targeting OsNIP3;1 via CRISPR/Cas9: A strategy for minimizing arsenic accumulation and boosting rice resilience

Arsenic (As) contamination in rice poses a significant threat to human health due to its toxicity and widespread consumption. Identifying and manipulating key genes governing As accumulation in rice is crucial for reducing this threat. The large NIP gene family of aquaporins in rice presents a promising target due to functional redundancy, potentially allowing for gene manipulation without compromising plant growth. This study aimed to utilize genome editing to generate knock-out (KO) lines of genes of NIP family (OsLsi1, OsNIP3;1) and an anion transporter family (OsLsi2), in order to assess their impact on As accumulation and stress tolerance in rice. KO lines were created using CRISPR/Cas9 technology, and the As accumulation patterns, physiological performance, and grain yield were compared against wild-type (WT) under As-treated conditions. KO lines exhibited significantly reduced As accumulation in grain compared to WT. Notably, Osnip3;1 KO line displayed reduced As in xylem sap (71–74%) and grain (32–46%) upon treatment. Additionally, these lines demonstrated improved silicon (23%) uptake, photosynthetic pigment concentrations (Chl a: 77%; Chl b: 79%, Total Chl: 79% & Carotenoid: 49%) overall physiological and agronomical performance under As stress compared to WT. This study successfully utilized genome editing for the first time to identify OsNIP3;1 as a potential target for manipulating As accumulation in rice without compromising grain yield or plant vigor.

Effects of different light intensity on leaf color changes in a Chinese cabbage yellow cotyledon mutant.

Leaf color is one of the most important phenotypic features in horticultural crops and directly related to the contents of photosynthetic pigments. Most leaf color mutants are determined by the altered chlorophyll or carotenoid, which can be affected by light quality and intensity. Our previous study obtained a Chinese cabbage yellow cotyledon mutant that exhibited obvious yellow phenotypes in the cotyledons and the new leaves. However, the underlying mechanisms in the formation of yellow cotyledons and leaves remain unclear. In this study, the Chinese cabbage yellow cotyledon mutant 19YC-2 exhibited obvious difference in leaf color and abnormal chloroplast ultrastructure compared to the normal green cotyledon line 19GC-2. Remarkably, low-intensity light treatment caused turn-green leaves and a significant decrease in carotenoid content in 19YC-2. RNA-seq analysis revealed that the pathways of photosynthesis antenna proteins and carotenoid biosynthesis were significantly enriched during the process of leaf color changes, and many differentially expressed genes related to the two pathways were identified to respond to different light intensities. Remarkably, BrPDS and BrLCYE genes related to carotenoid biosynthesis showed significantly higher expression in 19YC-2 than that in 19GC-2, which was positively related to the higher carotenoid content in 19YC-2. In addition, several differentially expressed transcription factors were also identified and highly correlated to the changes in carotenoid content, suggesting that they may participate in the regulatory pathway of carotenoid biosynthesis. These findings provide insights into the molecular mechanisms of leaf color changes in yellow cotyledon mutant 19YC-2 of Chinese cabbage.

Exogenous methyl jasmonate enhanced kenaf (Hibiscus cannabinus) tolerance against lead(Pb) toxicity by improving antioxidant capacity and osmoregulators.

In this study, the effects of exogenous methyl jasmonate (MeJA) on metal uptake and its ability to attenuate metal toxicity in kenaf plants under Pb stress were investigated. The experiment was conducted with five different MeJA concentrations (0, 40, 80, 160, and 320 μM) as a foilar application to kenaf plants exposed to 200 μM Pb stress. The results revealed that pretreatmen of MeJA significantly increased plant dry weight, plant height, and root architecture at all concentrations tested, with the most significant increase at 320 μM. Foliar application of MeJA at 160 μM and 320 μM increased the Pb concentrations in leaves and stems as well as the translocation factor (TF) from root to leaf. However, the bioaccumulation factor in the shoot initially decreased and then increased with increasing MeJA concentration. By increasing enzymatic (SOD, POD, and CAT) and non-enzymatic (AsA and non-protein thiols) antioxidants, MeJA pretreatment decreased lipid peroxidation, O2- and H2O2 accumulation and recovered photosynthetic pigment content under Pb stress. Increased osmolytes (proline, sugar, and starch) and protein content after MeJA pretreatment under Pb stress restore cellular homeostasis and improved kenaf tolerance. Our results suggest that MeJA pretreatment modifies the antioxidant machinery of kenaf and inhibits stress-related processes that cause lipid peroxidation, hence enhancing plant tolerance to Pb stress.
